Glukose ist die mit Abstand wichtigste Nährstoffquelle für Mikroorganismen bei biotechnologischen Prozessen. In nahezu 95 % aller Kultivierungen wird diese Kohlenstoffquelle eingesetzt. Die Kontrolle der Glukosekonzentration kann sowohl für den Prozessverlauf notwendig oder schlicht aus Kostengründen ratsam sein. Ein typisches Beispiel ist die Hochzelldichtefermentation von E. coli zur Herstellung rekombinanter Proteine. In der Produktionsphase dieser Fed-Batch Kultivierungen darf die Glukose-Konzentration Werte von etwa 1 g/l nicht überschreiten, um die Bildung von unerwünschten Metaboliten zu verhindern. Vollständiger Verbrauch der Glukose führt jedoch zu sofortiger Abnahme der Lebendzellzahl. Die Regelung des Substratstroms benötigt daher eine zeitnahe und verlässliche Überwachung der aktuellen Konzentration.
